Cr(VI) Reduction by Bacteria Isolated from Mangrove Sediments of Ngurah Rai Forest Park, Bali

Abstract

This study aims to isolate and characterize chromium-resistant bacteria from the mangrove sediments of the Ngurah Rai Forest Park to identify potential candidates for Cr(VI) bioremediation. Sediment samples collected from four stations were used for preliminary screening on nutrient agar supplemented with 100mg/ L of K₂Cr₂O₇, resulting in the isolation of ten bacterial colony types. Five isolates demonstrated initial tolerance to Cr(VI) and were subsequently subjected to minimum inhibitory concentration (MIC) testing and Cr(VI) removal assays. The MIC results showed that isolate A exhibited the highest resistance, maintaining growth at Cr(VI) concentrations up to 1500mg/ L, while the remaining isolates displayed lower tolerance thresholds. Cr(VI) removal efficiency decreased with increasing chromium concentrations; however, isolate A consistently demonstrated the highest reduction performance across all treatments. Molecular identification based on 16S rRNA sequencing confirmed isolate A as Staphylococcus sciuri strain TY-42. These findings indicate that S. sciuri isolate A possesses strong potential for application as a Cr(VI)-reducing bacterium in contaminated mangrove sediment environments. © 2025, Egyptian Society for the Development of Fisheries and Human Health.
