Aspirin or acetylsalicylic acid (ASA) has been used for more than a century an anti-inflammatory. However, aspirin has side effects on the stomach when used in the long term. Caffeine or 1,3,7-trimethyl xanthine is a major ingredient in coffee that has both antioxidant and oxidant properties. Aspirin and caffeine have properties as chelating agents that potentially produce free radicals. Ovalbumin is an albumin (also called albumen) in egg whites. Ovalbumin has a metal-chelating capacity, especially in sulfhydryl groups. Ovalbumin, aspirin, and caffeine can form the AOC complex (aspirin-ovalbumin-caffeine) as a potential antioxidant and potentially allow electron transfer. Electron transfer is a system that drives biological processes such as oxygen binding, respiration, photosynthesis, and detoxification. The complex of AOC has been used as a preventive and therapeutic treatment for various conditions associated with inflammation and free radicals. The mechanism of AOC action in blood circulation has not yet been studied. This study measured plasma aspirin levels after sub-chronic AOC administration for 30 days. Because aspirin in plasma binds to albumin, HPLC was used to determine plasma aspirin levels. Rattus norvegicus were divided into 3 groups: control group; aspirin group given aspirin 5 mg/kg BW for 30 days; and AOC group orally given AOC for 30 days. Blood sampling after treatment was taken through the heart after the rat was sacrificed. The results showed that untreated mice had very low levels of aspirin (0.17 nmol/ml), while in rats treated with aspirin at the rate of 5 mg/kg BW for 30 days, there was an increase of blood aspirin level up to 7.76 nmol/ml. The AOC (contains 318.96 nmol/ml of aspirin) administration group for 30 days showed an increased blood aspirin level up to 5.92 nmol/ml. These data indicate that the administration of the AOC complex contributes to an increase in blood aspirin levels. This suggests that the AOC complex can be absorbed in the digestive system and brought into blood circulation, so it is possible that the AOC complex works in the blood circulation. © 2018 Author(s).
